Cell and organ specific delivery of drug and gene loaded nanoparticles is one of the major promises of the technology. Potential candidates of target molecule are membrane proteins expressed abundantly and/or selectively in the target organ which will be retained on the surface or internalized after ligand binding. Therefore, measuring membrane protein expression will give a rational strategy for the discovery of cell and organ specific delivery pathways. We have developed a method measuring spatial distribution with quantitative proteomics from formalin-fixed paraffin-embedded tissues. In the Flagship, we shall characterize membrane protein expression and find pathways for the ocular tissue specific delivery nanoparticles.
